چکیده
The chemical structure of oligonucleotide analogues dictates the conformation analogue oligomers, their ability to hybridize complementary DNA and RNA, stability degradation pharmacokinetic properties. In a study aimed at investigating new featuring neutral backbone, we explored oligomers containing morpholino-peptide backbone bind oligonucleotides. Circular Dichroism studies revealed our interact with DNA, molecular modelling interaction responsible for complex stabilization.

Morpholino antisense oligomers: design, preparation, and properties.
Antisense promised major advances in treating a broad range of intractable diseases, but in recent years progress has been stymied by technical problems, most notably inadequate specificity, ineffective delivery into the proper subcellular compartment, and unpredictable activity within cells. Herein is an overview of the design, preparation, and properties of Morpholino oligos, a novel antisens...
متن کاملBacterial resistance to antisense peptide phosphorodiamidate morpholino oligomers.
Peptide phosphorodiamidate morpholino oligomers (PPMOs) are synthetic DNA mimics that bind cRNA and inhibit bacterial gene expression. The PPMO (RFF)(3)RXB-AcpP (where R is arginine, F, phenylalanine, X is 6-aminohexanoic acid, B is β-alanine, and AcpP is acyl carrier protein) is complementary to 11 bases of the essential gene acpP (which encodes acyl carrier protein). The MIC of (RFF)(3)RXB-Ac...
متن کاملSynthesis and G-Quadruplex-Binding Properties of Defined Acridine Oligomers
The synthesis of oligomers containing two or three acridine units linked through 2-aminoethylglycine using solid-phase methodology is described. Subsequent studies on cell viability showed that these compounds are not cytotoxic. Binding to several DNA structures was studied by competitive dialysis, which showed a clear affinity for DNA sequences that form G-quadruplexes and parallel triplexes. ...

متن کاملMCR-1 Inhibition with Peptide-Conjugated Phosphorodiamidate Morpholino Oligomers Restores Sensitivity to Polymyxin in Escherichia coli
In late 2015, the first example of a transferrable polymyxin resistance mechanism in Gram-negative pathogens, MCR-1, was reported. Since that report, MCR-1 has been described to occur in many Gram-negative pathogens, and the mechanism of MCR-1-mediated resistance was rapidly determined: an ethanolamine is attached to lipid A phosphate groups, rendering the membrane more electropositive and repe...
